Abstract
A review of the published literature on propoxyphene hydrochloride was undertaken to determine if the wide popularity of the drug is justified. Only double-blind studies were reviewed; 20 were identified in a bibliography of 243 articles. In the course of the review, deficiencies in the design, conduct, and reporting of the clinical studies were noted. The addiction liability of propoxyphene is low but several reports have demonstrated its abuse potential. The reviewed studies show that propoxyphene is not superior to codeine or aspirin in terms of analgesic effect. No definite statement as to relative incidence of side effects can be made. It appears that factors other than intrinsic therapeutic value are responsible for the commercial success of propoxyphene.
